What Is NAD+?
NAD+ (Nicotinamide Adenine Dinucleotide) is a coenzyme found in every living cell and is essential for life itself. It plays a central role in cellular energy metabolism, DNA repair, gene expression, and cellular signalling. Despite its fundamental importance, NAD+ levels decline significantly with age — by as much as 50% between the ages of 40 and 60 — making it one of the most compelling targets in longevity research.
Mechanism of Action
NAD+ functions as an electron carrier in the mitochondrial electron transport chain, facilitating the conversion of nutrients into ATP (cellular energy). It is also a critical substrate for sirtuins — a family of proteins often called "longevity genes" — which regulate gene expression, inflammation, and stress responses. Additionally, NAD+ is consumed by PARP enzymes during DNA repair processes.
